Neuroprotective Effects of Silibinin-loaded Nanostructured Lipid Carriers (NLCs) in Aluminum Chloride-induced Neurotoxicity

چکیده
Nowadays, aluminum is commonly believed to be one of the potent neurotoxins, being highly associated with neurodegenerative diseases like Alzheimer’s disease. Nanostructured lipid carriers (NLCs) have been studied as novel drug delivery systems. A better therapeutic response occurs when a bioactive compound is delivered via NLCs as compared to when it is administrated as a standalone. The purpose of this study was to scrutinize the neuroprotective and antioxidant properties of Silibinin-loaded NLCs (Sili-NLCs) against AlCl3-induced neurotoxicity in male mice. Silibinin encapsulated in NLCs was prepared by the emulsification-solvent evaporation method. After mice treating, biochemical tests were carried out to distinguish the oxidative damages. Histopathological evaluation was performed by H&E staining. The particle size, polydispersity index, zeta potential, and entrapment efficiency (%) of prepared Sili-NLCs were to be found 239.7±4.04 nm, 0.082±0.003, -16.33±0.15 mV, and 72.65±2.03 %, respectively. Our results showed that AlCl3 not only caused a significant cognitive impairment, but also led to an alteration in oxidative stress in the brain tissue. On the other hand, treatment with Sili-loaded NLCs in comparison with free silibinin significantly improved the AlCl3-induced neurochemical anomalies. Results of this investigation illustrated that Sili-loaded NLCs are potentially superior to silibinin in neuroprotective effects against AlCl3.
